    Although Caco-2 cell-based permeation assays are used to predict the oral absorption of drugs and phytochemicals in humans, they lack an important feature of the human intestine: the mucus layer on the luminal surface. We have developed mucus-layered cell model using Caco-2 combined with the mucus-secreting HT29-MTX cell-line. We confirmed the presence of a mucus layer on top of Caco-2/HT28-MTX monolayers by scanning electron microscopy and fluorescence microscopy. β-carotene uptake in the mucus-layered Caco-2/HT29-MTX model was clearly lower than in the standard model when β-carotene was carried by larger micelles, but not when carried by very small PC micelles, suggesting that the mucus layer inhibited β-carotene uptake by intestinal cells, probably by slowing the permeation of larger mixed micelles. The new mucus-layered model could also effectively detect the enhancement of β-carotene absorption by certain phospholipids.
  • 食品成分による脂溶性栄養機能成分の成体利用性調節に関する研究

    カロテノイドを含む脂溶性栄養機能成分は油脂に比べ消化・吸収(生体利用性)が非常に低い。本研究では,植物性食品に含まれるこれらの脂溶性栄養機能成分の生体利用性高める調理・加工法及び食品素材の開発のため,脂溶性栄養機能成分の消化・吸収に対する脂質や食物繊維等の食品成分の影響を解析することを目的とする。単純拡散に従うものと考えられていた小腸上皮細胞へのカロテノイドの取り込みにトランスポーターが介在している可能性が最近報告されてきたため、本年度は,トランスポーターの関与とこれまで得てきた研究結果との関連を中心に検討することにした。 1.カロテノイドの小腸上皮細胞への取り込みにトランスポーターが関与する可能性を小腸上皮モデル細胞Caco-2を用いて検討した。細胞表層のタンパク質を分解するため細胞をトリプシンで処理した後,ミセル可溶化β-カロテンの取り込みを調べたが,処理前後で有意な差は認められなかった。トランスポーターに対する特異的抗体を用いてβ-カロテンの取り込みへのトランスポーターの関与を検討したが,適切な実験条件の設定にいたらず,今後の課題として残された。 2.疎水性の高い物質を小腸上皮細胞から排泄する他のトランスポーターがカロテノイド取り込みに与える影響を検討するため,Caco-2細胞によるローダミン排泄のP糖タンパク依存性を調べたが,明確なトランスポーターの活性が見出されなかった。ヒト肝臓由来細胞ではローダミン排泄活性が明確に見出されたので,この細胞を用いることによってカロテノイドに対する排泄活性の有無を解析することが期待される。 3.Caco-2細胞を用いた透過評価系で,β-カロテンの透過に対するミセルリン脂質の影響を調べた。ホスファチジルコリンは細胞への取り込みに対して抑制的に働くことが分かっていたが,透過に対しては促進的に働くことを見出した。リン脂質は細胞の脂質代謝に影響することによって,透過を促進する可能性が考えられた。
