    Involvement of four different genes encoding cysteine proteinase in the rapid wilting of balloon flower (Platycodon grandiflorus) petals was investigated. Expression of PgCP1 was up-regulated just after the flower opening stage and the expression remained until the onset of wilting. On the other hand, PgCP3, PgCP4 and PgCP5, which were newly identified from petals at the stage just before wilting, were markedly up-regulated from the stage just before wilting and the wilting stage. The promoter analysis indicated the possibility that the regulation via ethylene-responsive regulatory element (ERE) is involved in the expression of PgCP1, PgCP4 and PgCP5 in the petals whereas other unknown factors might be involved in regulation of PgCP3.
  • Molecular mechanism of ethylene-induced senescence in carnation flowers

    In this research project, we have investigated the molecular mechanism of flowering and senescence of carnation flower, which is a typical ornamental showing ethylene-dependent senescence, and obtained the following results. (1) We revealed several groups of genes which are up- or down-regulated in petal cells during flower opening by a suppressive subtractive hybridization technique. The putative functions of the translational products were classified into several categories including transcription, signaling, cell wall modification, lipid metabolism, and transport. Furthermore, four cDNAs encoding xyloclucantransglucosylase/hy-drolase, three cDNAs encoding expansin and a cDNA encoding sucrose synthase were cloned from petals of opening carnation flowers. Real-time RT-PCR analyses revealed that respective genes are differently expressed during petal development (elongation, expansion and outward-bending), indicating regulated expression of these genes during this process. (2) We revealed the presence of two genes of an ACC synthase (DcACS1), designated as DcACS1a and DcACS1b in the genome of 'Light Pink Barbara' carnation for the first time. They differ mainly in the nucleotide sequences of introns. Moreover, we cloned genomic DNAs of these genes, including their promoter region, and introduced them into tobacco plants. In the near future, it will be possible to see whether these genes will induce climacteric ethylene production in tobacco flowers undergoing senescence. (3) We discovered a gene encoding glycine-rich RNA binding protein (DgGRP1), which has an ethlylene response element (ERE) in its promoter and is down-regulated during carnation flower senescence. This gene will be analyzed in the near future by comparing with cysteine proteinase 1(DcCP1) gene, which has an ERE in its promoter but up-regulated during flower senescence.

    1.Analysis of ethylene production in the long-lasting carnation 'White Candle' In senescing carnation (Dianthus caryophyllus L.) flowers, ethylene is produced from the gynoecium, and acts as a diffusible signal received by petals to induce autocatalytic ethylene production in the petals. We discovered cv. ‘White Candle (WC)' as a long-lasting cultivar, of which cut flowers produce ethylene only in trace amounts and have a long vase life. The low ethylene production in these flowers was due to low ethylene production in the gynoecium, accompanied by low accumulation of ACC synthase (DC-ACS1) transcripts. These findings further support the importance of ethylene production from the gynoecium in the senescence of carnation flowers. 2.Cloning and characterization of a cDNA for a novel carnation cysteine proteinase The wilting of carnation petals is caused by degradation of cell components, which leads to cell death during senescence of petals, mediated by some hydrolytic enzymes such as cysteine proteinase (CPase, DC-CP1), lipase and so on. We isolated a cDNA for a novel cysteine proteinase (DC-CP2) from carnation and compared its expression to that of DC-CP1 in senescing carnation petals. Furthermore, we isolated and characterized the nucleotide sequences of promoter region of both DC-CP1 and DC-CP2 genes. 3.Analysis of wilting and fading in senescing carnation petals We categorized senescence of carnation flowers was into two types ; the first is 'wilting' which is induced by the action of ethylene and the second is 'fading' which occurs independent on ethylene. We revealed that a cysteine proteinase (DC-CP1) and its inhibitor (DC-CPIn) were up-regulated in carnation petals undergoing wilting as well as fading.
