    The effects of long-term feeding of very low-carbohydrate diet on glucose tolerance and safety were studied in the genetic diabetic rats, Goto-Kakizaki (GK) rats. Forty-eight male GK rats (3 weeks of age) were divided by 16 into high-carbohydrate (HC) diet group, high-fat (HF) diet group, and very low-carbohydrate (LC) diet group and bred until all the rats died. The glucose tolerance was confirmed by feeding of LC diet temporarily, but was improved by a long-term feeding. In addition, the mean life span did not differ among the three groups. These results suggest that long-term feeding of LC diet improve glucose tolerance compared with HC and HF diets, but did not influence life span in GK rats.
  • Improvement of the digestibility of rare-sugars by nano-food-technology

    Rare-sugars have various physiological activities such as the decrease of body fat accumulation, the decline in serum glucose level, and the elevation of serum insulin level. Because of the low digestion ratio and the disposition different from glucose, the availability of rare sugars is limited. In this study, we attempted to improve the blood threshold of rare sugars by means of drug-delivery techniques. In order to apply for functional food we chose several safe, easily available, and inexpensive carriers and prepared the complexes of them with rare sugars. We assessed the effects of chronic feeding of the rare-sugars -carrier-complexes for rats to obtain the basic data on the safety of the complexes as nutritional substrates. No difference was found between the control group and the complex-fed groups in body weight gain, food efficiency, and most serum chemical levels. These results suggest that the rare-sugars -carrier-complexes have no dietary toxicity for rats.

    Although it has been difficult to supply a large amount of rare sugars, an inexpensive synthetic method on a large scale was recently established, leading to the reports on various physiological functions including regulation of blood glucose level and suppression of body fat accumulation. Meanwhile, it has been pointed out that rare sugars are difficult to utilize as foodstuffs by reason that they are excreted after the gut absorption more rapidly than typical monosaccharide. Therefore in present study, by applying drug delivery systems (DDS), we tried to bring the physiological functions of rare sugars into action efficiently by means of improving the stability in the body and prolonging the residence time. In addition, safety assessment methods for functional foods containing saccharides such as those exerted in the oral cavity were examined.
  • Sport nutritional study concerning inhibition of sarcopenia and osteopenia with aging in rats

    We examined the effects of a voluntary resistance exercise (climbing) together with high-protein snacks (pulse pattern) on bone mass and strength in rats given glucocorticoid-injections (2 mg/kg/day) as a model of age-related osteopenia. Wistar rats, 10 weeks of age, were assigned to low protein (15% of total food intake) or normal protein (20%) diet groups. Half of the rats were exercised daily during dark period (20:00-8 : 00 h). These groups were further divided into groups that received no snack or a pulse pattern high-protein snack (75% of dietary protein) during resting period. All groups were meal-fed at 8:30-9:30 h and 20:30-21:30 h and the snack was fed at 12:30-13:30 h for 8 weeks. Energy intake was approximately equal in all groups. The exercise groups were forced to climb a wire-mesh tower cage (20 cm diameter x 200 cm height) to drink water from a bottle set at the top. Bone weight and calcium content were increased by climbing with pulse pattern high-protein snacks. Climbing significantly increased bone maximum load and structural stiffness. However, no differences in bone mass or strength were observed between low and normal protein feeding groups. These results suggest that resistance exercise protects against the development of osteoporosis associated with aging. Pulse pattern high-protein snacks may enhance the effects of exercise.
  • 性周期と肥満に関する基礎的研究〜エネルギー代謝からの検討〜

    [目的] 月経周期がエネルギー代謝に影響を及ぼすことが知られている。本研究では、これまでに月経周期の長さと体脂肪率との間に正の相関が在ること、安静代謝および食事誘発性体熱産生が卵胞期に比べて黄体期で大きいことなどを明らかにしてきた。ところで、1日のエネルギー消費量を決定するものとして、安静代謝および食事誘発性体熱産生以外の要素として活動時のエネルギー消費量があり、1日のうちでかなりのウエイトを占めている。今年度本研究では、運動によるエネルギー消費量、すなわち運動中および運動後のエネルギー消費量が卵胞期と黄体期で異なるかを検討した。 [方法] 健康で月経周期が安定している19〜20歳の健常女性7名を被験者とした。実験の前日より被験者の食事を統一し、当日の早朝空腹時において最大酸素摂取量の80%程度の自転車漕ぎ運動を60分間実施させた。卵胞期および黄体期にそれぞれ運動前、運動中および運動後6時間にわたって、酸素消費量、二酸化炭素排出量および尿中窒素排泄量を測定し、これらを用いて運動中のエネルギー消費量、運動後の総エネルギー消費量、運動による運動後のエネルギー増加量を算出した。また、脂質酸化率の指標として、呼吸商を算出した。 [結果] 運動中のエネルギー消費量には卵胞期と黄体期で有意差を認めなかった。運動後の総エネルギー消費量、運動による運動後のエネルギー増加量は卵胞期に比べて黄体期で有意に大きかった(p<0.05)。安静時および運動中の呼吸商には卵胞期と黄体期で有意差を認めなかったが、運動後の呼吸商は卵胞期に比べて黄体期で有意に低くかった(p<0.05)。 [結論] 以上の結果から、黄体期は卵胞期に比べて運動後のエネルギー増加量と脂質酸化率が大きく、減量時の運動は黄体期でより効果的に作用する可能性が示唆された。
  • 牛脂高脂肪食がラット褐色脂肪組織脱共役蛋白と脳アドレナリンレセプターに及ぼす影響

    牛脂あるいは紅花油を添加した高脂肪食をラットに与えると、紅花油食に比べて牛脂食で褐色脂肪組織(BAT)の交感神経活性が低下し、BATの食餌誘発性体熱産生(DIT)が低下すること、その結果、牛脂食で体脂肪蓄積が増大することが報告されている。本報告では、牛脂食のDIT低下作用に、特異的熱産生蛋白質であるBAT脱共役蛋白(UCP)量の低下が関与しているか否かを検討した。 一方、牛脂食で交感神経活性が低下する原因として、視床下部及び大脳皮質のノルエピネフリン代謝回転速度(NEt)およびβ-アドレナリンレセプター(β-AR)結合能の低下が関与している可能性が報告されているが、詳細については不明な点が多い。本研究では、両群ラットの視床下部及び大脳皮質におけるβ-AR量および分布を明らかにするため、免疫組織化学的手法を用いて検討した。さらに、ノルエピネフリン合成の律速酵素であるチロシンヒドロキシラーゼ(TH)量が、牛脂食で低下しているか否かについても検討した。その結果、以下の知見を得た。 1.BATのUCP量は、紅花油食群に比べて牛脂食群で有意に小さかった(p<0.05)。 2.免疫組織化学的手法を用いて調べた、視床下部および大脳皮質におけるβ-ARの染色強度および分布には、両群間で差は見られなかった。 3.視床下部のTH量は、紅花油食群に比べて牛脂食群で有意に小さかった(p<0.05)。大脳皮質のTH量は、紅花油食群に比べて牛脂食群で低い傾向にあった。 以上のことから、牛脂食によるDIT低下作用には、BATのUCP量の低下が関与している可能性が示唆された。さらに、牛脂食で視床下部および大脳皮質のNEtが低下する原因として、TH量の低下が関与している可能性が示唆された。免疫組織化学的手法を用いて調べたβ-ARの分布については、明確な結果が得られなかった。
