In this study, we identified the SMN2 gene encoding an RNA helicase. The SMN2 is a target determinant of nuclear RNA exosomes, which are important for RNA quality control in the nucleus. SMN2 is involved in proper expression of the SMN1/RPS6 gene, which encodes an immune receptor for the HopA1 effector in Pesudomonas syringae, through the degradation of abnormal RNA generating from 3’region of the SMN1/RPS6 gene.

Elucidation of hypersensitive cell death mechanism by functional effectomics of Ralstonia solanacearum effectors

To elucidate a mechanism of hypersensitive (HR) cell death, the most important issue in plant immunity. In this study, we screened for the Ralstonia solanacearum effector and identified clone 42 that suppresses HR-like cell death induced by constitutive active form of MEK2 (MEK2DD). Agrobacterium-mediated expression of clone 42 in Nicotiana benthamiana resulted in chlorosis as well as increased expression of PR1. Infection of Agrobacterium expressing clone 42 in N. benthamiana leaves showed significant reduction compared to the control expressing GFP. These results suggested that clone 42 was recognized by the host. Taking into account, we conclude that the suppression of HR-like cell death by clone 42 is due to the reduced infection efficiency of coinfiltrated Agrobacterium expressing MEK2DD.

Mitogen-activated protein kinase (MAPK) pathways have a pivotal role in innate immunity signaling in plants. Arabidopsis MEKK1, a MAPK kinase kinase, constitutes specific pathway with MKK1/MKK2 and MPK4. This pathway has an important role in regulation of defense gene expression in response to microbe associated molecular patterns recognition. However, loss of MEKK1 results in constitutive defense responses accompanying severe dwarfism and temperature-dependent cell death. To elucidate the complex mechanism underlying the mekk1 phenotypes, we performed genetic screening to isolate suppressor mutant of the dwarf and defense phenotypes. With the aid of the MutMap analysis, we identified the candidate of causal gene for the mutant. Genetic analysis by producing double mutant with mekk1 or mpk4, smn1 mutation suppressed the dwarf and defense phenotypes of these mutants. These results showed that SMN1 positively contributed to the phenotypes.

To identify possible regulator of MEKK1, we performed yeast two-hybrid screening using MEKK1 protein as bait. We found two putative E3 ligases which contain U-box domain, named PUB25 and PUB26 as novel MEKK1-binding proteins. Characterization using pub25/ 26 double mutant suggested that PUB25 and PUB26 positively contribute MEKK1 function.
