    本年度は、新生仔豚4頭を用いて、脳組織の還流後の固定方法を変更し、それが免疫組織染色に適するものかを調べた。その結果、以前の固定法よりも変更後の固定法のほうが、免疫組織染色では反応が安定することが明らかとなった。また、KB染色での正常神経細胞の数を調べ、水素ガスにより、正常神経細胞が増加するかを調べたところ、水素ガスでは吸入時間が6時間でも、軽症負荷では正常神経細胞が増えることが分かった。
  • 妊娠母体炎症が惹起する児の脳-身体の双方向性制御機能の破綻

    妊娠中の母体感染が児の脳発達及びエネルギー代謝に与える影響を明らかにするため、妊娠炎症曝露モデルを用いて影響を受ける児脳領域と、それによって変動する行動異常を明らかにする。特に視床下部と自律神経系には特別に焦点を当て、妊娠中の炎症による視床下部への影響と末梢組織(肝・膵・腸管・脂肪組織など)の変化を網羅的に解析し詳細に評価する。これにより、妊娠中の慢性的母体感染が如何なる機序(例えば、神経、内分泌・生理活性物質など)で、脳-末梢臓器の連関を破綻させるのか、そしてそれは将来の肥満を基盤とする生活習慣病の発症に如何に影響するのかを明らかにする。

  • New therapy for neonatal resuscitation using hydrogen gas

    新生仔豚16頭を用いて、低酸素虚血負荷を行い、無治療群と水素ガス吸入群の2群にわけて、負荷後6時間まで人工呼吸器管理を行い、近赤外光時間分解装置による脳循環酸素代謝変化、脳波計による脳波測定、及びバイタルデータの記録を行った。また、負荷後6時間で、臓器還流固定を行い、脳組織の病理組織評価を行った。その結果、無治療群では、異常脳波を3頭で認め、それらの脳組織における正常神経細胞は認めなかったが、異常神経細胞は1視野あたり10個程度認めた。水素ガス吸入群では、3頭で異常脳波を認め、それらでは、正常細胞を1視野あたり50個程度認め、異常細胞は1頭のみ1視野当たり10個認めた。以上の結果より、水素ガス投与下では、異常脳波を伴う場合でも正常神経細胞が生存可能となることが示唆された。
  • 母子分離モデルから紐解く発達障害発症に関わる脳領域

    最終年度では、前年度 (2020年度)に確認された光遺伝学による扁桃体中心核の活性化が攻撃行動を惹起することについて、その信頼性を高めるための追試験と組織学的解析を行った。方法は、アデノ随伴ウイルスを用いて脳定位手術にて9週齢のSDラット雄の扁桃体中心核にチャネルロドプシン-2を発現させ、その直上にLEDカニューラを導入した。2020年度に問題となった再現性を改善すべく導入部位の微調整及びアデノ随伴ウイルスの力価を減少させたところ、非常に高い再現性でLEDでの光刺激(λ=470nm)による攻撃行動の惹起に成功した。また組織学的解析を行ったところ、チャネルロドプシン-2の発現が扁桃体中心核に限局していること、更に光刺激は扁桃体中心核を主に活性化させていることをc-Fos発現で確認できた。最も懸念されるのは、すぐ近傍に存在し攻撃性に関与する事が数多く報告されている扁桃体内側核も同時に活性化されて攻撃行動を惹起してしまった可能性であったが、少なくとも我々の条件下ではチャネルロドプシン-2を扁桃体内側核に発現させ光刺激をくわえても攻撃行動が惹起されなかった。これらの結果は、扁桃体中心核自体の過剰活性が過度な攻撃性(あるいは暴力性)と非常に強く関係している事を示すものであり、2019年度に認められた母子分離されたラット仔の過剰な攻撃行動の一端は扁桃体中心核の過剰な活性状態にその原因があることを支持する結果である。扁桃体中心核の過剰な活性がもたらす攻撃性は’Predatory aggression (略奪的攻撃性)’との関係が指摘されており、それはヒトにおける非行・暴力を伴う犯罪行為に繋がる事が示唆されている。従って本研究課題から見出された結果は母子分離、すなわち幼少期の育児放棄のような不遇な養育環境は将来的な反社会的行動に繋がる根本となる攻撃性に関わる脳領域の発達に影響を与えうることを示唆するものである。
  • Bidirectional signaling mechanism between the intestinal microbiota and brain: Molecular basis of obesity induced by stress.

    Numerous studies reported on the effects of early life stress on brain and body development. However, little is known about how it alters the intestinal bacterial flora. The new vistas for understanding the brain-gut interactions related to the gut microbiota provide further insights to understanding the developmental mechanism of brain-gut interactions. In this study, we examined the gut microbiota of a maternal separation model in rats and found changes in composition particularly related to lifestyle-related diseases and inflammation. Furthermore, this animal model showed an increased expression of inflammatory cytokines released by microglia in the brain during early life. These results suggest that early life stress alters an inflammation-prone gut microbiota, which may cause inflammation in the brain.
  • Establishment of a new neonatal resuscitation strategy by monitoring cerebral hemodynamcis and oxygen metabolism as an indicator

    We measured cerebral haemoglobin oxygen saturation (ScO2) and cerebral blood volume (CBV) using three-wavelength near-infrared time-resolved spectroscopy in a 5-day survival asphyxiated piglet model. We investigated how changes in ScO2 and CBV during the resuscitation period after hypoxic-ischaemic (HI) insult reflected histopathological brain injury on day 5 after insult. Changes in CBV after HI insult were correlated with histopathological brain injury but changes in ScO2 were not. Clinical study, we examined CBV and ScO2 during 15 min after birth between healthy term neonates delivered vaginally (VD, n=13) and those delivered by elective cesarean section (CS,n=18) using TRS. CBV was significantly higher in the VD group than in the CS group in the 4 min after birth (p<0.05) but not thereafter. There was no significant between-group difference in ScO2 during the transition period.
  • Influence of Early Life Stress on the Brain Development

    Previous studies have suggested that BDNF, which induces synaptic development, induces CYP46 which promotes the biosynthesis of 24-hydroxycholesterol. In the present study, we investigated the possibility that 24-hydroxycholesterol could be a biomarker for synaptic development. Specifically, we evaluated the potential of 24-hydroxycholesterol to serve as a biomarker of synaptic development using repeated maternal separation (MS), a model of preterm birth stress. The results showed that the expression of CYP46 in the hippocampus was reduced by MS. These results suggest that early life stress may have reduced CYP46 expression levels via a reduction in BDNF. Therefore, stress during development may act on the brain to induce a decrease in 24-hydroxycholesterol, suggesting that 24-hydroxycholesterol could be used as a marker for synapse formation.

    The relationship between the modification status of the sex-determining gene Sry on the Y chromosome and the expression of sex-differentiation-related factors was investigated. As a result, it was revealed that Sry expression was different among the commonly used mouse sublines, and that there were spatiotemporal and lateral differences in Sry expression in healthy male individuals. The organ culture system and internal normalizing genes required for validation of Sry expression were also established. Moreover, it was revealed that Amh acts on the Mullerian duct by infiltrating the tissue from the testis toward the mesonephros.
  • Relationship between hyperbilirubinemia and cerebral oxygen metabolism in newborn infants.

    Neonatal hyperbilirubinemia is the unique physiological phenomenon in human. Its biological significance depends on the role of bilirubin as a radical scavenger during neonatal period, when the brain oxygen metabolism develops rapidly. We validated the methods for measurements of both liver glucuronidation and cerebral oxygen consumption in human neonates. We found that serum conjugated bilirubin and bilirubinⅨαare useful for assessments of liver glucuronidation two weeks after birth, because the secretion of conjugated bilirubin is stable at that time. Further, we investigated relationships between cerebral Hb oxygen saturation (ScO2) by near-infrared spectroscopy and Hb oxygen saturation of the internal jugular vein or femoral artery in children with congenital heart diseases, found ScO2 is useful for assessing cerebral oxygenation in neonates.

    The purpose of this study was to establish an objective index of maternal-infant interaction and to elucidate the mechanism of attachment formation between mothers and their children. Using a two-channel portable near-infrared tissue oxygen monitor with near-infrared spectroscopy, blood circulation in the prefrontal cortex of the mother and infant simultaneously was measured during the three occasions of "Separation," "Caring," and "Breastfeeding. We examined the correlation of oxygenated hemoglobin in mothers and infants by moving the mother's data time with respect to the child (cross-correlation), and examined the time lag at which the absolute value was maximized. The histogram figure showed a parametric distribution and the largest distribution between 0 and 0.3. We also examined cross-correlation ranks by three occasions and found that there was a tendency to show more stronger correlation during breastfeeding than during caring.
  • The Influences of maternal separation on neural circuit formation during brain development equivalent to preterm infants in human

    In this study, we analyzed the influences of maternal separation (MS) during brain development equivalent to preterm infants in human using rat model. Our results showed that MS during brain development increased aggressive behavior and reduced the behavior related to social recognition after maturation. In addition, we found that reduction of various factors (VGAT, Neuroligin-2) related to inhibitory synapse in the amygdala and the medial prefrontal cortex of mature rat exposed to MS. Excitatory/inhibitory imbalance disrupts normal function in these regions, which leads to cause excessive aggression and abnormality of sociability. Our study also revealed these influences of MS are not irreversible, but rather can be recovered depending on nurturing environment after MS.

    This study was carried out to examine the effects of early postnatal maternal separation stress on the developmental crosstalk between the brain and peripheral energy-related organs. Our study can obtain insight understanding etiological mechanisms of life style-related diseases, such as obesity. Maternally separated rats showed abnormal alterations in neurotrophic factor expressions, their signal transduction systems and synapse structure protein phosphorylations. Furthermore, maternal separation stress affected various enzymes that involves in lipid metabolism. Examination on gut microbiota in these animals showed a disturbance in microbial composition leading to cause abnormal metabolism and/or nutrition. Our study showed that maternal separation stress during early postnatal life can affect the functions of both brain and peripheral organs that causes obesity in later life.
  • Effect of BDNF on fatty acid homeositasis during brain development

    It is suggested that lower dietary intake of nutrients such as fatty acids during the development is related to the vulnerability to the on-set of psychiatric disorders. However, brain fatty acid homeostasis in the neonatal period has not been fully understand. In this study, we tried to elucidate the brain lipid homeostasis in this period. As a result, it was found that saturated fatty acids are actively desaturated in neonatal brain, that BDNF (Brain-derived neurotrophic factor) is involved in liver SCD-1 activity and can participate in systemic fatty acid homeostasis, and that BDNF induces the expression of HPA axis-related protein.
  • Relationships between the regulation mechanisms of sex differentiation and the epigenetic status in the developing gonad

    We examined the relationship between regulation mechanisms of gonadal sex differentiation and disorders of sex development (DSD). In the DSD line mice (YPOS), inappropriate spatiotemporal expression of sex determine gene Sry and sex differentiation related genes in the developing gonad cause different phenotype from the same genetic background. We found new regulatory mechanism about sex differentiation related gene dmrt1 using non-coding region of dmrt1 gene. The epigenetic status of this region was non-methylated both in the testis and ovary in adult mice. Furthermore, it became clear that, there were asymmetries in the expression patterns of dmrt1 and differentiation process of Sertoli cells in the developing chick gonad.
  • The effects of hydrogen gas inhalation for hypoxic-ischemic encephalopathy in piglets.

    The aim of this study is to evaluate the effectiveness of free radical scavenger (edaravone, EV) combined with hypothermia (HT) for hypoxic-ischemic (HI) brain injury, and the safety of hydrogen gas inhalation in neonatal piglets. The following results had been reported. At first, there was a positive linear correlation between changes in cerebral blood volume and low-amplitude aEEG duration during 12 h after the insult in the no-therapy group, but a negative linear correlation between these two parameters in the HT group. Secondly, both HT and HT+EV groups showed lower histopathological cerebral damages compared to that of no-therapy, especially in gray and white matter. However, HT+EV did not show lower damages compared to HT. Lastly, hydrogen gas inhalation had no effects on circulatory and metabolic activities, especially cerebral hemodynamics and oxygen metabolism, and cortical electrical activity. Therefore, hydrogen gas is likely to be used safely in neonatal medicine.
  • Analysis of molecular mechanisms involved in the susceptibility to statins on cancer cells

    The anti-proliferative effect of stains on cancer cells has received widespread attention, since it has been reported that the statins may exhibit an anti-cancer effect in addition to their cholesterol-lowering effect. However, what type of cancer cell statin is effective for is unclear. Our previous study identified E-cadherin as a candidate factor determining the susceptibility to statin. To demonstrate that E-cadherin actually determines the susceptibility of cancer cells to statin, we examined if the regulation of E-cadherin expression has an influence on the susceptibility of cancer cells to statin in the present study. When E-cadherin gene (CDH1) was inserted into the statin-sensitive cells which had no E-cadherin, the half maximal inhibitory concentration (IC50) increased 3.7-fold. On the other hand, E-cadherin knockdown in the statin-resistant cancer cells increased their susceptibility to statin. These results strongly suggest that E-cadherin is a statin resistance factor.

    In these studies, we found that combining the cerebral blood volume (CBV) with amplitude-integrated EEG (aEEG) is a more effective guide to control hypoxic/ischemic (HI) insult in a newborn piglet model than aEEG alone, to produce a consistent degree of survivable neuropathological damage. Next, we found that an early CBV increase and longer low-ampitude EEG after insult indicates severe brain injury. Finally, we revealed that Edaravone (a free radical scavenger) reduced histologic neuronal damage related to HI loading in our piglet HI model. However, we also found the histopathological damage, especially kidney tissue. We have to consider the systemic impact by Edaravone.

    Physical exercise preconditioning is becoming generalized in patients with hypertension to reduce a risk factor of cerebrovascular diseases Based on such therapeutic framework, we examined whether physical exercise preconditioning prior to the intracranial-hemorrhage can attenuate the cerebrovascular damage in rat hippocampus or not. Present study revealed that physical exercise preconditioning prior to intracranial hemorrhage can attenuate neuronal damages in both young and aged animals. However, precise etiological mechanisms involved in the preconditioning effects on the brain is unknown at present. Further studies are needed to clarify this phenomenon.
  • Risk of learning deficit and mental disorder by prolonged maternal separation

    Present study showed that prolonged maternal separation influenced formation of neuronal circuit in the CA1 region of rat hippocampus and hippocampus-dependent learning ability after weaning. In addition, prolonged maternal separation caused the reduction of BDNF-ERK signaling and hyperphosphorylation of GluR1 via CaMKII in the rat hippocampus. Given that these factors are closely related to synaptic formation, functional synapses are commenced to be formed around postnatal days 7-10, these alterations possibly disrupt normal functional acquisition of brain development. Our findings suggest that prolonged maternal separation disturbs the factors related to synaptogenesis during early brain development, which might be in part responsible for abnormalities of formation of neuronal circuit and learning deficit in later life.
  • Alterations of synaptic transcytosis induced by ethanol exposure: non-vesicular synaptic transport in the rat

    Neuronal tracer of HRP is considered to be transported independently at synapse. To investigate this transport, the solitary nucleus was observed at the electron microscopic level after WGA-HRP injection into the rat vagus nerve. Animals receiving injection of WGA-HRP with Rab3A-siRNA or alcohol treatment revealed apocrine-like structures of terminals which contained HRP reaction product and indented into secondary neurons. Finally, these structures were observed to be separated completely from axon terminals. Quite similar structures were found in the posterior pituitary in animals receiving same injections into the hypothalamus or alcohol treatment. The apocrine-like structures of hypothalamic terminals showed to contain HRP reaction product and large dense core vesicles, and be frequently separated into the vascular space. The present study apparently confirmed the existence of synaptic transport of proteins which was quite different from previous vesicular synaptic transport.

    This study was carried out to examine the effects of early postnatal maternal separation stress on the brain development, biological properties of adipose tissue, and the interactions between them. Wistar rats were separated from their mothers in early life. The level of various neurotrophic factor ligands/receptors in the cerebral cortex and cerebellum showed abnormal time-dependent fluctuations. Given that these factors play important roles in brain development, it can be speculated that the altered expression of these factors induced by maternal separation may interrupt normal brain development and ultimately lead to functional disruption. Furthermore, there was a significant change in the level of various mitochondrial energy-expenditure molecules. Given that these molecules influence the mitochondrial metabolism, our study indicates that early postnatal maternal deprivation can influence the fate of adipose tissue proliferation, presumably leading to obesity later in life.

  • Neurotrophic factors and cognitive function after alcohol intoxication in mice model

    Here we investigated the effects of ethanol (EtOH) and acetaldehyde (AcH) on behavior,and mRNA levels of nerve growth factor (NGF), brain-derived neurotrophic factor (BDNF) and c-fos at 30 min after EtOH in Aldehyde dehydrogenase 2 knock (Aldh2-KO), Apolipoprotein E knockout (ApoE-KO) and wild type (WT) mice. Eight-arm-radial maze and Morris-water maze showed that 1) 2.0 g/kg EtOH impaired spatial memory in all three strains, while lower doses did not have consistent memory effects, 2) a probe trial(drug-free state) showed a sustained memory impairment in mice that had received 2.0 g/kg EtOH previously, regardless of strain, 3) ApoE-KO mice learned more slowly than WT mice, and the Aldh2-KO mice learned more quickly.Real time PCR showed that 1) EtOH increased c-fos mRNA expression in ALDH2-KO (1 and 2g/kg) and WT (2.0 g/kg), 2) 2.0 g/kg of EtOH decreased ChAT mRNA in Aldh2-KO mice, while acetylcholinesterase mRNA expression was increased in both strains, 3) acute EtOH failed to alter the mRNA levels of NGF and BDNF and 4)Aldh2-KO mice showed a distinctly lower level of ChAT mRNA and a higher level of NGF mRNA versus WT mice. Our observation suggests that high EtOH and AcH can alter cholinergic markers and this effect did not modify the levels of NGF and BDNF.

    Notch signaling, related with maintenance of Stemness in culture, is well known to play an important role in neuroprotective and proliferative properties. Therefore, alcohol exposure showed to activate the Notch pathway. As results, Notch signaling related receptors, such as Platelet-derived growth factor (PDGF), Brain-derived neurotrophic factor (BDNF) and Nerve growth factor (NGF), were indicated to be strongly expressed. However, there are significant lacunae about formation of neuronal network under expression of these receptors. Particularly, changes of neuronal membranes, axonal transport and synaptic transmission should be studied. In this point, a study of synaptic transport of protein might provide biological solution for psychiatric disorder of alcoholism.
  • Effects of maternal care on brain development : molecular mechanism of mother-infant interactions

    Wistar male rats were separated from their mothers for 3 hours each day between PND10 and 15 (MD group). Nurotrophin ligand and receptor expressions were analyzed. The expression of BDNF/TrkB in MD rats was enhanced in both mRNA and protein levels. In contrast, no significant difference in NGF/TrkA expression level was observed between these groups. On the other hand, OMgp expression was elevated in MD animals. Abnormal fluctuations in those trophic factors cause organic changes, and ultimately lead to functional disturbance.

    本研究では, 器官形成期におけるエストロゲン様化学物質の曝露が, 如何にして不可逆的な生殖障害を引き起こすのか, そのメカニズムの一端を解明することを試みた. 本研究により, 視床下部-下垂体-性腺軸一連の機能障害メカニズムにおいては, より中枢側の責任が大きいこと, また, 精巣ライディッヒ細胞における側鎖切断酵素P450scc遺伝子は, ヒストン脱アセチル化を引き起こすことにより遺伝子発現が低下し, アンドロゲン産生能の低下に至ることが推察された. 興味深いことに, 天然エストロゲンではP450scc遺伝子のヒストン脱アセチル化が引き起こされず, エストロゲン様内分泌攪乱物質と天然エストロゲンでは, ヒストン修飾のレベルで作用機序が異なることが示された.

    新生児期の脳酸素代謝の特徴は、早産児は正期産児と比較し脳血液量、酸素代謝量が低く、在胎週数と共に増加する。しかし低酸素虚血性脳症や脳室内出血を来たす病態では、出後早期には脳の循環自動調節能が破綻し、脳血液量が増加し、酸素消費量も低下する事が示唆された。特に低酸素虚血性脳症のエネルギー代謝障害時には、軽症例では酸素消費量が減少するが、重症例では脳血流上昇後に脳浮腫が進行し虚血が進行すると考えられた。
  • Molecular biological approach to explore the cause of brain damage after prenatal ethanol exposure

    To identify genes and factors which are responsible for brain alterations after prenatal ethanol exposure, we examined expression of genes and determination of neurotransmitters or neurotrophic factors in ethanol-exposed rat brains. Immunohistochemical study using anti-5-HT antibody revealed that prenatal ethanol exposure resulted in a reduction in the number and defects of migration of serotonin-positive neurons in the dorsal and medial raphe nuclei of rat fetuses. In ethanol-exposed rat fetuses, amounts of both 5-HT and its metabolite (5-HIAA) were decreased as well. RT-PCR analysis revealed a reduction of sonic hedgehog (Shh) expression in the brains of ethanol-exposed rat fetuses. Because Shh is related to the fate of 5-HT neuron, we concluded that prenatal exposure to ethanol impairs the development of 5-HT neurons, and this might have resulted in the reduction of Shh expressions. Therefore, the reduction of 5-HT neurons seen in the raphe persisted into adulthood, and prenatal ethanol-exposed rats showed anxiety-like behavior. In ethanol-exposed rat cerebellum, BDNF mRNA was decreased but NGF and its receptor (Trk A) mRNA expression were increased. Also, reduction of OMgp and GDNF mRNA expression was observed in the hippocampus and the septum nucleus, respectively. Thus, it is established that ethanol can affect expression of certain neurotrophic factors and their receptors. We monitored the intracellular calcium concentration after glutamate stimulation in ethanol-treated astrocytes derived from the hippocampus of neonatal rat (Day 3), and revealed that the glutamate-reactive cell number was decreased compared to non-ethanol-treated astrocytes. Since the calcium concentration in astrocytes might be related to maturation of synapse, the result suggests that ethanol may affect development of the neural circuit via a calcium signaling cascade in astrocytes.
  • Involvement of reward system in acquisition and maintenance of alcohol independence.

    Wistar rats were exposed to a relatively high daily dose of ethanol at postnatal day 10-15 by placing them for 3 h/day in a chamber containing ethanol vapor. These ethanol-treated rats (blood concentration : approximately 430 mg/dl) were found to have slightly but significantly fewer pyramidal cell neurons. The present observations indicate that pyramidal cells in the hippocampus may be vulnerable to a relatively high dose of ethanol exposure during this short period of early postnatal life. In additional experiment, the effects on the frontal cortex and hilar neuron numbers, observed as a result of such short periods of ethanol treatment, appeared to be transitory. With respect to colocalization of taurine and glial fibrillary acidic protein immunoreactivity in mouse hippocampus, induced by short-term ethanol exposure, quantitative analysis revealed that the merged area in ethanol-treated mice was twice that (71.6% vs. 35.8%) of control mice. Since taurine is involved in various neuroprotective functions, the present observations suggest that the expression of taurine immunoreactivity in reactive astrocytes after ethanol exposure might play an important role in neuroprotective processes. Additional analysis was made to investigate the effects on expression of neurotrophic factors. The expression levels of brain- and glia-derived neurotrophic factors (BDNF and GDNF) in the hippocampus were analyzed by RT-PCR analysis. This study revealed that chronic ethanol consumption does not result in significant changes in expression levels of BDNF and GDNF mRNAs. However, it was characteristic in myelin-associated glycoprotein that expression levels of oligodendrocyte myelin glycoprotein (OMgp) mRNA were increased. This appears to be consistent with the neuropathological changes in alcoholics, which have relationship with demyelination.
  • Effects of Maternal Deprivation on Brain Development in Rats

    We have established the experimental animal (rat) model of maternal deprivation (MD) in human by separating pups from mothers for 3 h/day during postnatal days (PND) 10-15. Effects on body and pituitary weights The body weight of MD rats showed no significant difference at PND16, 20 and 30, whereas at PND60 it was high compared to age-matched control. On the other hand, pituitary weight was low at any time points of observation. Effects on brain weight The brain weight of MD rats showed no significant difference at any time points of observation. Corticosterone concentration During the separation period, blood corticosterone concentration of MD rats was significantly higher (51ng/dL) than that of controls (32ng/dL). After separation, it was declined to 40 ng/dL. Stereological analysis (physical disector) There were no significant difference in total number of pyramidal cells in CA1-3 regions, and granule and hilus neurons in the dentate gyrus of MD rats compared to controls. Additionally, numerical density and the regional volume of MD rats showed no differences compared to controls. Real-time PCR analysis of BDNF mRNA MD rats had less BDNF mRNA in the hippocampus at PND16, whereas at PND 30 and 60 they had much BDNF mRNA than age-matched control animals.
  • Effects on neuronal formation of the hippocampus in developmental abnormalities

    The effects of prenatal X - irradiation and ethanol exposure on neuronal development of the hippocampus were investigated in the present study. At six weeks of age in the rats the laminal structure of the hippocampus was deranged with exposure of 0.6 Gy or more. Pyramidal cells in the CA3 region were more susceptible than those in the CA1. Abberant mossy fiber terminals were observed in the stratum onence of the CA3 region with exposure of 0.3 Gy or more. On the other hand, exposing neonate rats to high dose of ethanol affected Purkinje cell numbers in the cerebellum. The period between 4 and 9 day of age was an extremely vulnerable period during which the Purkinje cells were particularly susceptible to the effects of a high dose of ethanol. However, a similar level and duration of ethanol exposure commencing after 10 day of age had no significant effect on the cell numbers. In contrast to granule and pyramidal cells numbers, neurons in the hilus region were indicated to be particularly vulnerable to the effects of a high dose of ethanol exposure during early postnatal life.
