  • On molecular mechanism to produce phenotypically adaptive variation among genetic clones of ants

Insects often exhibit mechanical movements, and if genes govern behavior, it is expected that individuals with the same genes will have the same behavioral threshold. We observed initial and conditioning-induced changes in behavioral thresholds in the Icariidae, clones of the same nest worker with a common gene set, and examined genomic differences between individuals and found no differences. To ascertain whether this diversity is regulated by DNA methylation epigenetics, we analyzed differences in methylation status among individuals with different behavioral thresholds and found no differences in methylation status within the target range.
  • Evolution of army ant syndrome: lessons from non-army ant doryline ants
Japan Society for the Promotion of Science:Grants-in-Aid for Scientific Research
    Date (from‐to) : 2016/04 -2019/03
    Author : Ito Fuminori

Colony composition, prey menu, reproductive cycle, caste differentiation and foraging behavior were investigated for non-army doryline ants in southeast Asia. Colony size varies from 20 in most species to more than 1000 workers in a few species of Cerapachys. At least seven species containe polygynous colonies. All species of Lioponera showed non-cyclic reproduction whereas other species showed cyclic reproduction. Prey specialization varies species to species: some Cerapachys species fed on various different kind of arthropod while Lioponera and some other genera accepted ant brood only.
  • Differences in behavioral traits among supercolonies of the Argentine ant Linepithema humile
Japan Society for the Promotion of Science:Grants-in-Aid for Scientific Research
    Date (from‐to) : 2013/04 -2016/03
Author : Inoue Maki; ITO Fuminori

We examined the behavioral traits among the four supercolonies introduced into Japan. Each L. humile supercolony demonstrated different behavioral traits: JM and KB exhibited strong aggressiveness toward a foreign worker, whereas KC avoided a foreign worker but exhibited more foraging activity than the others. The KA supercolony infrequently attacked a foreign worker and explored food. We analyzed mitochondrial DNA of L. humile workers from native Argentina populations. Sequencing revealed the six clades: the first clade included JM, KB, and the populations collected across Argentina; second, KC, the eastern region and Sndes region; third, only the eastern region; fourth, Tokyo colony. Geographically genetic structure was not detected and thus the species have been expanding associated with the development of the trade along Palana River and railways.

Colony composition of 15 species of genus Gnamptogenys was studied in Southeast Asia. 13 of 15 species were reproduced by dealated queens without worker sexual reproduction. In G. menadensis, both dealated queens and mated workers laid eggs. Former colonies showed monogyny while the later colonies were polygyny. In G. bicolor, large ergatoid queens, dealated queens or mated workers reproduced. The colonies were always monogyny. Large ergatoid queens had been found only in East Java, where no alate queens were collected. Mated workers also reproduced in East Java. In the other populations, mated workers and dealated queens laid eggs. Body size of alate/dealate queens varied among populations.

The Argentine ant, Linepithema humile, is highly polygynous and possesses a social structure, called supercolony (SC). In this study, different populations widely distributed across the continent share the same haplotype, whereas small SCs are pettily distributed in the restricted areas. The genetic analysis suggested that gene flow among SCs is very limited, and SCs are closed reproductive units. Furthermore, the workers from small SC showed their escape behavior and whereas the workers from massive SC attacked the workers from other SCs in groups. Our findings suggested that a massive SC displace other SCs and form the huge colonies across the globe

Effects of Argentine ants on native animals except for ants were investigated in and around Hatukaichi-si, Hirosima Prefecture. Effects of the Argentine ants on ground living arthropod animals were obscure, however, the density of a Japanese lizard was reduced in the invaded area. The nest density of tube nesting wasps in ant foraged branches was lower than that in ant excluded branches in argentine ant invaded area. Host specialist myrmecophilous animals are remarkably affected by the invasion of Argentine ants.

We analyzed the expanding dynamics and impact of Argentine ants (Linepithema humile), an invasive alien species in Japan, on native ant community using Bayesian statistical models based on a long-term monitoring data taken from 2000 to 2012 in Hatsukaichi-city, Japan. The results clearly showed that the Argentine ants in high density decreased the density of native ant species, while the ecological factors controlling on their expansion could not detected.

We confirmed that Argentine ants are expanding their distribution along the coastal areas of Pacific Ocean side in Japan, though we succeeded in eradication of one small population in Yokohama using Z-(9)-Hexadecenal as well as a common myrmecocide. To prevent the invasion of RIFA into Japan, we developed methods for identification (using DNA bar coding) and detection (using the parasitoid wasp Microplitis croceipes) which can readily be used even by quarantine officers. In the surveys in Taiwan, USA and Argentina, we found that polyploid workers rapidly increased in the countries where RIRA had recently invaded. Using Diacamma sp. which is useful for molecular biology, we studied the sex determination mechanism in ants and found some intriguing expression patterns of genes such as feminizer.

To reveal the role of ants as prey in terrestrial ecosystem, prey menu and prey preference of frogs and ant-eating arthropods, and effects of ants on survival and growth of a Japanese treefrogs were investigated. Hyla japonica and Microhyla ornate fed on many ants, however, they show remarkable preferences : they did not prefer to eat ant species with strong defensive weapons and small sized species. When we kept Hyla japonica by giving only ants, survivorship and growth rate were significantly different among ant species.

In this research on Formica yessensis which exhibits a supercolony on the coast of Ishikari, Hokkaido, We confirmed the presence of a chemosensory sensillum which was previously discovered in workers of a phylogenetically close ant Camponotus japonica. Whereas the sensillum responded even to hydrocarbons of nestmates, the response was less intense than that to hydrocarbons of workers from other colonies, indicating that this organ of F. yessensis retains the function of nestmate recognition. Within the supercolony, the response of the sensillum was significantly correlated with the geographic distance of the analyzed nests, suggesting that there is a little antagonistic relationship even between the nests which do not show any aggressive behaviors. The presence of the antagonistic relationship between the supercolonial nests was also suggested by a mathematical analysis of the behavioral data which was obtained in the outdoor observation of F. yessensis workers. Moreover, the analysis of microsatellite DNA demonstrated that the kin relatedness within the supercolony was as low as that between different colonies. Although the intranidal kin relatedness was a little higher, the standard deviation was high, indicating that the nestmates were composed not only of kins but also of non-related workers and queens. These results suggest that the kin selection is not an important factor to maintain the supercolony. Probably, one of more important factors is the strong wind which often blows flying alates to the sea and accordingly offers advantages to the post-mating queens which abandon the flight and enter the natal or neighboring nests. The relatively homogeneous habitat on seaside dune may be another key factor which facilitated the development of polygyny and the reduction of internidal antagonism on the Ishikari Coast.

The distribution of the Argentine ant, Linepithema humile, in Japan, and effect of the ant on the Japanese biota were investigated. Main results are as follow. 1.The distribution of the Argentine ant was investigated in 65 cities or towns along the Seto Inland Sea, western Japan. Our results include all available information of their distribution in Japan until 2005. Argentine ants have invaded Aichi Pref. (Tahara-shi), Hyogo Pref. (Kobe-shi), Hiroshima Pref. (Hiroshima-shi, Fuchu-cho, Hatsukaichi-shi, Ono-cho, Otake-shi), and Yamaguchi Pref. (Iwakuni-shi, Yanai-shi). The distribution range of Argentine ants in each area was limited to a few km except for the area in and around Hatsukaichi-shi. 2.The distribution of Argentine ants in urban parks around Hatsukaichi city was investigated from 2000 to 2005. The number of parks invaded by the ant increased from 27 to 41 during six years. 3.The Argentine ant invasion has remarkably reduced species richness of resident ants. Among 11 common epigeic species, nine species were vulnerable to Argentine ants, whereas Paratrechina sakurae and Camponotus vitiosus appeared less affected by this invasion. 4.Prey composition of the Japanese treefrog Hyla japonica was investigated in area invaded by Argentine ant Linepithema humile and area not invaded by the ant. In both area, ants were the most common taxa among prey animals in young treefrogs, constituting 82.4% of numeric proportion and 49.9% of volumetric proportion, and 42.8% of the numeric proportion and 25.7% of the volumetric proportion in invaded and uninvaded area, respectively. Among ants, the Argentine ant was most abundantin invaded area while Tetramorium tsushimae was the most common prey item in uninvaded area.

Morphology and functions of exocrine glands in Asian tropical and subtropical ants were investigated. Main results were as follow. 1. Morphology of some novel exocrine glands of ants was investigated and described. 2. Food menu of the Japanese treefrog Hyla japonica was investigated and ants were main prey items. Function of defense substances of some ants was examined by using Hyla japonica and other real predators. 3. In Diacamma sp., the occurrence of worker policing toward individuals with developing ovaries was demonstrated Cuticular substances might be possible key stimuli for the policing behavior. 4. In temperate regions, older eusocial Hymenopteran females with annual life cycles tend to dominate younger females, a behavior demonstrated by many Polistes. However, in queenless ponerine ants and primitively eusocial tropical wasps (perennial/tropical), a younger female can be dominant and occasionally takes over from the older, most dominant reproductive female, the alpha. Assessment of individual age based on cuticular substances might be one of the possible mechanisms of these behaviors. Adaptive significance of these patterns in their life history was investigated by using an inclusive fitness model. 5. In tropical forests, ants frequently consume fruit pulps around seeds of vertebrate-dispersed plants. Seed cleaning behavior by omnivorous ants was observed in West Java. Field experiments on seed cleaning by the myrmicinae ant P. plagiaria revealed that the anti-fungal effect was not merely caused by removal of fruit pulps, and that seed cleaning behavior by P. plagiaria inhibited the development of spores and hypha of fungi. We suggest that the workers apply anti-fungal substances to the fruit pulp and seed surface. Such substances might be produced in mandibuler glands.

Social structure, symbionts and parasites of ant colonies, and local ant fauna were investigated in Indonesia and Malaysia. Ant fauna in Bogor Botanic Gardens, west Java, Indonesia was reported. Even though the Botanical gardens located in the center of city, we collected 216 ant species. Social structure of ponerine ants was investigated, and we found clear relationship between social structure and habitat characteristics : gamergate species (reproduced by mated workers)were common in disturbed area while ergatoid species (reproduced by wingless queens) were common in forests. Furthermore, in Gnamptogenys, gamergate species was collected in humid tropics (West Malaysia and Sumatera) while ergatoid species was found in seasonal tropics (east Java). These relationships between social structure and habitat condition seems to be explained by the frequency of opportunity of colonial fission. Relationship between social structure and symbiont biology was examined for Myrmecina. Morphological and behavioral specialization of symbionts is apparently related to social structure of host ants : ant species with ergatoid queens only have very specialized symbionts.

The purpose of this research project is to reveal the nutritional ecology of social insects on Sulawesi Is. In Indonesia and on Malay Peninsula in Malaysia. The social Insects such as termites and ants are abundant, and the influence of their activities for aphids, wood-feeding cockroaches and Passalid beetles in the tropical forest ecosystems are very strong. Therefore, the field surveys for the nutritional ecology of termites, ants and also of wood-feeding cockroaches, aphids and Passalid beetles were carried out in the northern part of Sulawesi Is. on SEpt.1995, and also done in the Malay Peninsula on March 1999. We focused the surveys to evaluate the influence of ecological factors such as the habitat, food resources, competitive species etc. on their form, behavior and social structure.

コロニー内の産卵女王数は、A.silvestrii,H.nubatama,B.chinennsisは多女王制であったが、他の8種は単女王制コロニーしか採集できなかった。

This project aims to clarify long term dynamics of insect populations in Java and Sumatra, Indonesia. We established three major study sites under different climate conditions, (1) Padang, central Sumatra, has a typical humid tropical climate without a distinct alternation of wet and dry seasons. Annual rainfall exceeds 5000mm, (2) Bogor, west Java, located at the eastern fringe of the humid tropical climate, has an annual rainfall of 4075mm with more seasonality than Padang, (3) Purwodadi, east Java, has a distinctly seasonal climate. Annual rainfall is > 2000mm with 2-6 consecutive months of < 100mm rainfall. Major study species included phytophagous lady beetles (Coleoptera : Coccinellidae : Epilachninae), tortoise beetles (Coleoptera : Chrysomelidae : Cassidinae), and the banana skipper Erionota thrax (Lepidoptera : Hesperiidae). Seasonal fluctuation in abundance and mortality factors of these insect populations has been documented in detail in relation to the changes in climatic conditions and the action of natural enemies. Regular field census, including mark-recapture of adults and construction of life tables, was carried on the lady beetles (Epilachna emarginata in Bogor, E.vigintioctopunctata in Bogor, Padang and Purwodadi), and on the tortoise beetles (Aspidomorpha miliaris and A.sanctaecrucis in both Bogor and Purwodadi). The census periods for these studies have covered from 2 to 4 years, and some of them are still going on. Eggs, larvae, and pupae of the banana skipper were collected at a regular interval from the field and reared in the laboratory to examine faunal diversity and intensity of actions of parasitic wasps and flies. Laboratory rearings were also executed to clarify the survivorship and reproductive schedules of E.vigintioctopunctata collected from Bogor, Wonogiri (central Java), Sukarami (central Sumatra).
