    初年度の研究においてトロンビン受容体PAR1を欠損するマウスは、高脂肪食による肥満とインスリン抵抗性は野生型マウスと同定に生じるものの、加齢に伴うインスリン抵抗性が特異的に抑制されていることを見出した。これを受けて、PAR1拮抗薬を用いた薬理学介入が加齢に伴うインスリン抵抗性の発症に対して予防及び改善効果を発現するか否かを解析した。野生型マウスにおいて若齢期(8週齢)から80週齢まで長期間にわたりPAR1拮抗薬を飼料に混じて投与し、加齢に伴うインスリン抵抗性の発症予防効果を検討した。80週齢時のインスリン感受性および耐糖能を評価したところ、薬物投与群と薬物を投与しなかった対象群とに有意な差が認められなかった。さらに、高齢期(50週齢)から80週齢までPAR1拮抗薬を投与し、一旦成立したインスリン抵抗性および耐糖能障害を改善するかを検討した。80週齢時のインスリン抵抗性および耐糖能は、薬物投与群と薬物を投与しなかった対象群と差が認められなかった。 PAR1欠損マウスの脂肪組織の解析を行った。野生型マウスの脂肪組織(精巣上体周囲・腎臓周囲・肩甲骨間皮下・臀部皮下)は、加齢に伴い体重に占める湿重量比が増大するが、PAR1欠損マウスのそれは緩やかであった。 PAR1を全身性に欠損させることによる脂肪組織の減少と代謝改善効果(インスリン感受性や耐糖能)に関わる遺伝子発現変化を明らかに知るため、脂肪組織(腎臓周囲脂肪組織)・肝臓・腓腹筋のマイクロアレイ解析を行った。現在詳細を解析中である。

The Rho-Rho kinase pathway plays an important role in the peripheral localization of ppMLC and actin filament formation during the initial phase of the thrombin-induced endothelial barrier disruption. 3. The coagulation factor XIa induces Ca2+ signal via proteinase-activated receptor 1(PAR1) and L-type Ca2+channel in vascular smooth muscle cells. 4.     Cerebral vasospasm is one of critical complications that develop after subarachnoid hemorrhage and influence vital and neurologic prognosis of the patients. The present study aims to develop innovative and ultimate strategies for the treatment of cerebral vasospasm with thrombin receptor as a therapeutic target, based on the understanding of the central mechanism of pathogenesis of vasospasm. As a result, the present research elucidated the mechanisms underlying (1) impairment of desensitization of thrombin receptor PAR1, (2) thrombin-induced endothelial barrier disruption and (3) thrombin-induced store-operated calcium influx. All these aspects play a critical role for the development of cerebral vasospasm. The findings of the present study therefore consolidated the basis for the development of a novel therapeutic strategy with PAR1 as a therapeutic target for the prevention and treatment of cerebral vasospasm.

    The present study yielded a new mechanism for endothelial barrier dysfunction. By using an animal model of pulmonary hypertension, a thrombin receptor antagonist was found to effectively prevent the development of pulmonary hypertension and prolonged the survival. The contractile responsiveness of pulmonary artery was found to be enhanced in pulmonary hypretension. As a result, thrombin receptor antagonist is suggested to be potentially useful as a new strategy for treatment of pulmonary hypertension. Proposals of Japanese and international patent for the invention of new treatment for pulmonary hypertension has been submitted.

    The present study elucidated the role of proteinase-activated receptors(PARs) in the early phase of the development of atherosclerotic vascular lesions. Thrombin increased endothelial permeability by two distinct pathways by activating PAR1 ; one is mediated by Gα13, and independent of Ca^<2+>, but dependent of Rho kinase-mediated MLC di-phosphorylation ; other is mediated by Gαq, and independent of MLC di-phosphorylation, but dependent on Ca^<2+>.

    動脈硬化や血管形成術後の血管局所では、収縮性の亢進・攣縮を認めることがある。病変局所の平滑筋細胞や浸潤・増殖細胞において、収縮性亢進に関係する種々の酵素・蛋白質の発現変化が起こっている、と一般に考えられているが、その直接証拠は少ない。人工血管を作製し、これらの酵素・蛋白質の遺伝子を人工血管壁構成細胞に発現させることによって、収縮性の亢進・攣縮におけるその役割の直接証明が可能となる筈である。本年度は線維芽細胞のcell lineであるNIH3T3細胞を用いる事によって、等尺性収縮を測定可能な人工血管中膜モデルを作成した。さらにNIH3T3細胞に高効率に遺伝子導入できるベクターを開発し、CPI-17の過剰発現による線維芽細胞の収縮性の変化を検討した。CPI-17を過剰発現させた細胞とコラーゲンゲルを用いて作製した血管中膜モデルのリング標本は、PKCの活性化剤であるPDBuの投与により、10%FBSによる収縮の2-3倍の張力を発生した。一方、ベクターだけを発現させたコントロールではPDBuの投与により、むしろ基線以下に弛緩した。また、PKCによるリン酸化部位である38番目のThrをAlaあるはGluに変異させたCPI(CPI38AおよびCPI38E)では、PDBuによる収縮は得られず、コントロールと同様に弛緩した。CPI-17を過剰発現させた細胞のPDBuによる収縮は、PKC in...

    細胞間接触により増殖を停止したブタ大動脈由来培養内皮細胞からサイクリン依存性キナーゼ阻害因子p27^の新たなアイソフォーム(p27^)をクローニングした。p27^のがん抑制因子としての機能を、細胞および個体レベルで明らかにすることを目的として、核移行、細胞周期制御及び分解抵抗性に関わる構造を明らかにした。また、自然高血圧発症ラット(SHR)と正常ラットの大動脈における2つのアイソフォームの発現解析から、組織増殖性とアイソフォーム発現の相関を、個体レベルで明らかにした。1.p27^は、領域153-168が有意な核移行に必要な配列であることが明らかになった。この領域には塩基性アミノ酸は一残基(K168)しか含まれない。不足する塩基性アミノ酸に代わり疎水性アミノ酸が機能を果たすという点で非典型的な2分裂型核移行シグナルであることを明らかにした。2.GFP発現系を用いて細胞増殖に及ぼす影響を解析し、p27^の増殖抑制作用を明らかにした。増殖抑制には、p27^と共通のN末端領域が必要である。3.6週齢SHRの大動脈にはp27^が、正常ラットではp27^が、主なアイソフォームとして発現する。週齢を重ねると、SHRにおいてもp27^発現が優位になる。正常ラット大動脈を培養...

    A novel isoform of p27^, a cyclin-dependent kinase inhibitor, has been isolated from a cDNA library of porcine aortic endothelial cells. The N-terminal 162 amino acid of the new isoform was identical to those of p27^, while it contained a unique 18 amino acid C-terminus.     正常細胞の増殖制御において最も重要で、生理的な役割を果たす細胞間接触による増殖停止に関して、そのメカニズムを明らかにするために、培養ブタ大動脈内皮細胞を用いてp27^の発現調節機構を明らかにした。内皮細胞を低密度(コンフルエント密度の25%)及び高密度(80%)の二つの異なる密度で植え付け、異なる時間経過で細胞同士の接触による増殖停止を誘導することにより、p27^蛋白質の発現変化と細胞周期進行との時間的相関を明らかにした。静止期の細胞を植え付け直すと、25%及び80%植え付け共に細胞は同調してS期に入り、20時間後にS期のピークに達した。その後、25%植え付けの場合は96時間後に、80%植え付けの場合は48時間後に細胞周期から外れた。核におけるp27^蛋白質の発現は、細胞周期に入る際に消失し、再び静止期に戻る際に回復した。このp27^蛋白質の発現変化は、Kip1 mRNAの発現変化を伴った。p27^蛋白質の分解活性は、細胞周期に入る際に増加したり、細胞周期から外れる際に低下することはなかった。Kip1 mRNAの安定性は増殖期に低下し、細胞周期から外れる時期及びコンフルエント(静止期)の時期に上昇した。Nuclear run-on assayによりKip1遺伝子の転写活性はKip1 mRNAの発現レベルと関連すること...

    血管内皮細胞cDNAライブラリーからサイクリン依存性キナーゼ阻害蛋白質p27^の新規分子種をクローニングした。新規分子種は、従来種とは異なるC末端を有するスプライスアイソフォームである。細胞抽出液を用いた分解アッセイを行ったことろ、従来種は増殖期の細胞抽出液によって1時間以内に分解されたのに対し、新規分子種は抵抗性であった。このことから新規分子種を、p27^(degradation-resistant isoform of p27^)と命名した。新規分子種を緑色蛍光色(GFP)との融合蛋白質として培養細胞に発現させると、従来種と同様に核へ局在することが明かとなった。GFP蛍光顕微鏡画像解析により核局在の程度を定量評価する方法を確立し、従来種の変異体を解析したところ、従来種の核移行シグナルは2分裂型であり、領域153〜166が核移行に必要最小限の領域であることを明らかにした。ところが、従来種と新規分子種の共通領域では核への特異的集積が消失した。このことから、新規分子種に特異的なC末端領域にも核移行に必要は配列があり、新規分子種の核移行シグナルも2分裂型であることが示唆された。GFP蛍光を指標に、フローサイトメーターを用いて、組換え蛋白質発現陽性及び陰性細胞の増殖を評価する方法を確立した。すなわち、遺伝子導入後各時点で、総細胞数を計測し、発現...

    血管平滑筋の形質転換に伴うαトロポミオシン・アイソフォーム変換の調節に関与する情報伝達機構を明らかにする事を目的として今年度の研究を行った。1. 平滑筋細胞を培養すると、培養3〜5日目を最大に、一過性に非平滑筋型αトロポミオシンの発現が増加した。非平滑筋型アイソフォームの発現増加に一致して、BrdUの取り込みが、培養3日目を最大に一過性に増加した。2. コンフルエントの細胞を、2日間血清を含まない培地で培養し、再び血清刺激を行うと、刺激1.5時間で、非平滑筋型αトロポミオシンの発現が増加した。血清濃度1%から増強作用が認められ、3%で最大効果に達した。この時、非平滑筋型アイソフォームの発現は刺激前の約2倍に増加した。3. 血清再添加による非平滑筋型アイソフォーム発現増加は、H-7処理により完全に抑制されたが、wortmanninは有意な抑制作用を示さなかった。4. phorbil 12,13 dibutyrateは、血清刺激と同様に、刺激1.5時間で非平滑筋型アイソフォーム発現を増加させた。1nMから増強作用が認められ、100nMで最大効果に達した。この時、非平滑筋型アイソフォームの発現は刺激前の約2倍に増加した。5.     本研究は、平滑筋特異的スプライシング因子を同定するために、培養平滑筋細胞を用いて、平滑筋細胞の形質変換に伴うトロポミオシンのアイソフォ-ムの変換調節機構を明らかにすることを目的として行った。培養細胞には、ラット大動脈平滑筋細胞の初代培養を用いた。10%胎児牛血清を含む培地で培養し、コンフルエントに達した細胞を無血清培地で2日間培養して、実験に用いた。α-トロポミオシンアイソフォ-ム発現の解析は、逆転写-PCRにより行った。ラットα-トロポミオシンのエクソン1、2、3、4に特異的なプライマ-を作製し、平滑筋細胞から精製した総RNAを1μg用い、RT-PCRによりトロポミオシン発現を解析した。1.無血清培地で培養した平滑筋細胞を、再び血清で刺激すると1.5時間以内に、エクソン3を含む非平滑筋型アイソフォ-ムの発現が増加した。エクソン1、エクソン2の発現は変化しなかった。血清濃度1%から増強作用が認められ、3%で最大効果に達し、エクソン3の発現は刺激前の約2倍になった。2.血清によるエクソン3の発現増強は、1-10μM H-7の前処理により完全に抑制された。wortmanninは、0.1μMの濃度において、不完全な抑制作用を示した。3.フォルボ-ル12,13ジブチレ-トで平滑筋を刺激すると、エクソン3の発現が増加した。1nMから増強作用が認められ、100nMで最大効果に達し、エクソ...
